Release date: 2008-04-14 US scientists develop a new single-molecule DNA sequencer Researchers at Helicos BioSciences in the US recently said they have developed a new type of sequencing device, single-molecule DNA sequencers. The instrument is able to "read" a single base of single-molecule DNA without the need to amplify the DNA strand, reducing the chance of introducing errors in the process, making sequencing of the entire genome easier.

The new method adopted by Helicos BioSciences is called "synthesis sequencing": the virus M13 is used as a test object, first its genome is cut into small pieces, and an enzyme is used to attach a short DNA tag to the end of each fragment, where appropriate. The location anchors the DNA fragment. DNA replicase and bases or base pairs with fluorescent tags are then added. When the fluorescent DNA forms a strand, each newly added base pair is photographed with a camera. This method is capable of detecting the fluorescence of a single base.
This method avoids the cumbersome amplification process and will greatly reduce the time and cost of sequencing. Helicos BioSciences estimates that the new instrument can sequence a person's genome in eight weeks at a cost of $72,000, while the instrument itself is worth $1.35 million.
The new discovery has undoubtedly taken a solid step toward the $1,000/person grand genome sequencing project. The first author of the paper, Timothy Harris, believes that this goal will be achieved within five years. ——Shanghai Medical Device Industry Association
