Chlorophyll meter use steps and precautions

The chlorophyll meter is an instrument that can be taken directly into the hand and brought to the field for chlorophyll content determination in the chlorophyll meter. Because the instrument is small and can be held at any time, it is called a chlorophyll meter. The principle of measurement of a handheld chlorophyll meter is to measure the transmission coefficient in two wavelength ranges to determine the relative amount of current chlorophyll in the blade. The handheld chlorophyll meter has a measurement range of 0.0-99.9 SPAD. A lot of chlorophyll content is now shown by the SPAD value, because there is a certain correlation between the SPAD value and the chlorophyll content, because the chlorophyll content of the leaves can be known by measuring the SPAD value. In addition, in plant research, we often use the living area leaf area instrument to measure the plant leaf area, which is an important step in our crop yield estimation.

The specific workflow of the chlorophyll meter (SPAD502) is as follows: two LED light sources emit two kinds of light, one is red light (peak wavelength 650nm), one is infrared light (940nm), two kinds of light penetrate the blade, hit the receiver The optical signal is converted into an analog signal, the analog signal is amplified by the amplifier, converted into a digital signal by the analog/digital converter, and the digital signal is utilized by the microprocessor to calculate the SPAD value and display it on the display, and automatically store it in the memory. . The internal flow can be expressed as illumination system -> receiver -> amplifier -> analog / digital converter -> microprocessor -> display.

The SPAD value of plants has a certain proportional relationship with the chlorophyll content. The higher the SPAD value, the better the growth condition. When using the chlorophyll meter, the method is relatively simple. You can get the SPAD value of the plant by holding the chlorophyll meter and then clamping the measuring head of the instrument to the ends of the blade and pressing the measuring head.

The specific operation is:

(1) During the calibration process, the measuring head does not clamp the sample, the two LEDs emit light sequentially, and the received light is converted into an electrical signal, and the ratio of the light intensity is used for calculation.

(2) After the sample is clamped by the measuring head, the two LEDs emit light again, and the light transmitted through the blade hits the receiver and is converted into an electrical signal, and the intensity ratio of the transmitted light is calculated.

(3) The values of steps 1 and 2 are used to calculate the SPAD measurement, which is the current chlorophyll relative content of the clamped sample leaves.

When using the chlorophyll meter, you need to pay attention to the following points: 1. The reading verification card can only be used in the inspection mode. 2. Only a random reading check card (the same serial number as the machine) can provide an accurate value. 3. The reading verification card cannot be used outdoors, and cannot be used in direct sunlight, high temperature and high humidity. 4. When the measured value is different from the value on the reading check card, it cannot be corrected by inputting the compensation value. 5. Do not touch the glass surface of the reading verification card. If it is dirty, use a wet, soft cloth. 6. The reading verification card should be placed in the accessory bag for protection and should not be placed in a high temperature or high humidity environment.

Snacks

Snacks usually refer to the food eaten outside the time point of three meals a day. Under normal circumstances, in people's life, except that three meals a day are called dinner food, the rest are called snacks. Canned fruit is a healthy snack.
