The reporter learned from the Hefei Institute of Physical Science of the Chinese Academy of Sciences that Zhang Zhongping, a researcher at the Institute of Intelligent Micro-Nano Devices of the Institute of Intelligent Research, hired a team of researchers to make breakthroughs in tumor cell detection and accurate surgical navigation. Research results have recently been published in the international chemical journal ACS Nano, and have applied for national invention patents. Uncontrolled growth and immortalization of tumor cells requires the catalysis of highly active telomerase. The researchers designed gold nanoparticles as the carrier and designed a spherical nucleic acid probe with a large amount of specific double-stranded DNA on the surface. Under the catalysis of telomerase, the probe can release the fluorescent dye into the cytoplasm, so that the tumor cells emit red fluorescence, so as to achieve the visual detection of tumor cells. Through the change of the fluorescence signal, more than a dozen representative tumor cells and normal cells were accurately distinguished, the malignant degree of tumor cells was identified, and the live tumor imaging, naked eye visualization, and tissue section identification of the mouse tumor were performed. The results show that this spherical nucleic acid probe is an ideal tumor-guided surgical navigation contrast agent. The current tumor surgery is mainly based on the doctor's experience and subjective judgments, and lacks effective means to distinguish between tumor and normal tissues. In this study, the spherical nucleic acid contrast agent allows the tumor cells to “glowâ€. With the accuracy of their localization and high sensitivity, they can help doctors to determine the location and boundary of the tumor from the cellular level, so that the lesion can be accurately excised and the surgical risk can be reduced. At the same time, the postoperative recurrence rate and mortality of the tumor are greatly reduced. It is reported that the team will further develop animal and clinical trials for precise surgical resection of tumors, and eventually develop contrast agents for clinical tumor precise surgery and promote industrial applications.
